What it is
Semax is a heptapeptide, a synthetic analog of the ACTH(4-7) fragment extended with a Pro-Gly-Pro tail. That tail is functional: it resists the enzymes that would otherwise break the peptide down in minutes, which is what makes an intranasal dose useful. It is sold as a sealed, original Russian-format 0.1% nasal solution.
How it works
A single intranasal dose raises BDNF and activates its TrkB receptor in the hippocampus, the same growth-factor signaling tied to learning and neuron maintenance, while modulating dopaminergic and serotonergic tone (Dolotov et al., Brain Res 2006). It does not work by flooding the brain with dopamine the way caffeine or modafinil do, which is why most users describe clarity rather than a wired feeling. The plasma half-life is only minutes, but the central effects run several hours.
Dosing
The 0.1% solution delivers roughly 50 micrograms per drop. A common nootropic protocol is 2 to 3 drops per nostril, taken 1 to 3 times across the day. Start at the low end to gauge your response before increasing.
Dose in the morning or early afternoon. Effects begin in 5 to 15 minutes and run 4 to 8 hours. Late dosing is not associated with the sleep disruption stimulants cause, but most users keep it to daytime out of habit.
